Effects of GABA on Action of GRPergic Neurons in Exocrine Secretion of Isolated, Perfused Rat Pancreas

Abstract
Although GABA is synthesized in pancreatic ¥â-cells, its physiological role in pancreatic secretion is completely unknown at the present time. Recently, we firstly reported that exogenous GABA enhanced pancreatic secretion evoked by electrical field stimulation (EFS) in the isolated rat pancreas1. Since intrapancreatic GRP neurons exert a stimulatory role in pancreatic secretion2, this study was aimed to see whether GABA affects the action of the GRP neurons on pancreatic secretion. GABA (10 ¥ìM) elevated GRP (30, 100, 300 pM)-stimulated exocrine secretion in the isolated, perfused rat pancreas. GABA (10 ¥ìM) enhanced pancreatic secretion evoked by EFS, which was significantly inhibited by either an anti-GRP serum or atropine (2 ¥ìM). The GABA-enhanced EFS-evoked pancreatic secretion was further reduced by a simultaneous infusion of the anti-GRP serum and atropine.
The results indicate that GABA enhances pancreatic secretion evoked by activation of GRP neurons in the isolated, perfused rat pancreas. The GABA action is resulted from elevation of action of GRP which is released from the GRP neurons during the activation. Supported by Korea Research Foundation (F00052), 1999.
